Semen Detection Testing
Semen detection tests are used to visualize the presence of semen in stains on items, particularly personal items and articles of clothing. The test is a very reliable presumptive test that detects the high levels of an enzyme called acid phosphatase present in semen, even after a vasectomy. If the presence of semen is detected, the client may request additional testing to determine whether his or her DNA profile matches the DNA found in the stains or is excluded, meaning that the stain contains DNA from another individual.
DNA analysis assumes the presence of sperm in the sample. Positive presumptive tests for the presence of semen will be followed by microscopy to visualize sperm cells, which contain DNA. No profiles will be produced if there is no sperm in the semen stain. This can occur if the man had a vasectomy or is infertile because of low or no sperm production.
Once semen and sperm cells are detected and a decision is made to proceed with DNA testing, profiles will be generated from the DNA in the stains. At that time, a buccal (cheek) swab sample will be requested from the client to determine whether their DNA profile matches the DNA isolated from the stain.

Semen Detection on personal items (per item) Clothing or other items should be stored in new, clean paper bags or paper envelopes, never in plastic bags. Drying does not interfere with the test. Moisture destroys the sample over time. The test is not destructive and items will be packaged and returned to you unless you give us other instructions. Large items may require additional shipping fees. Call for details. DNA profiles supporting positive semen detection (optional)Additional optional testing would include generating DNA profiles. The additional testing usually requires that at least 2 DNA profiles be produced. Call for details. Home Semen Detection Kit "Infidelity testing" kits by CheckMate (www.getcheckmate.com) are available for home use. These kits are for presumptive detection of semen. The kits are easy to use and reliable for their intended purpose. Each kit is actually a double kit that tests up to a total of 10 items.
